Malachite green and magnesium chloride act as selective agents, combined with high osmotic pressure and low pH to effectively inhibit non-target competitive organisms. Soya peptone provides a source of nitrogen and potassium phosphate acts as a buffer. Simply dissolve 25 capsules per 1 Liter of distilled water. Water must be 37°C or warmer to completely dissolve gelatin capsule. Agitate. Sterilize by autoclaving for 15 minutes before use. Buffer is set for 7.0 ± 0.2 at 25°C using a 10mM solution of Tris/Tris HCl. pH will vary depending on temperature.
